Spoked rims require a tube. Really the only bikes that have spoked rims these days are dirtbikes(for obvious reason) and cruisers(for looks). BMW's spoked wheel has a patented technology that does not require a tube.

Yup.

And those cruisers generally use the same tire whether the rim is spoked or not. Wonder how they do that.. ;)

I just can't imagine taking a chance with a tire that has had a nail through it. I would have the tire changed right away.

Many tyres are made so they can be TL or tube

If that's a tubeless tire on a cast rim made for tubeless tires I'd just plug it on the bike and used it up and have done so a few times unless the nail is within about an inch of the edge and might not plug safely.

As far as putting a tube in the tire here is what Metz says:

Is it possible to fit a tube into a tubeless tyre?
Herewith we certificate that METZELER tubeless tires can be used with tubes on the admissible standardized rims. The allowable top speed is 130,43 mph for the "H"- tires and 142,86 mph for the "V"- and "Z"- tires You are bound to use the same product concerning the tubes and the tires on your motorcycle.
Meaning that METZELER-tires have to be used with suitable METZELER-tubes. Existing regulations of the motorcycle manufacturers have to be observed.

I plugged one like new rear Dunlop touring tire and used it another 20k miles before I traded the bike in with quite a bit of tread left on it. I wouldn't run a plugged tire on a mc unless I plugged it myself.

I mix a lot of tires and have rarely had a problem with it but I use my own judgement on what I'll mix. Here's the tires I've run on my last two bikes, a couple GS500's:
