This road has been listed here before but here is a little more information on it from "BCRider99". From Vancouver it can be ridden in a day but to get the most out of it scenic wise you can easily lengthen it into a two day jaunt.

Duffey Lake Loop

This is a superb loop that never grows old.

The best section is from Horseshoe Bay to either Lytton or Cache Creek. Traffic levels drop noticeably once past Whistler and then again past Pemberton. Pemberton to Lillooet is deservedly the most famous biking road in BC. You can find better pavement elsewhere but nothing beats the curves, remoteness, sparse traffic and unique scenery of the Duffey Lake Road !!

For some reason, #12 south from Lillooet to Lytton is little known. But it's a real sleeper. #12 is an excellent biking road with a constant barrage of curves, limited traffic and splendid scenery. Alternatively, the Marble Canyon route to Cache Creek is almost as good but adds about an hour to the trip making the day slightly long. I usually leave Marble Canyon for the Kamloops Loop (the section between Cache Creek and Lytton is fairly forgettable in any case so not a lot is missed).

The route through the Fraser Canyon (Lytton to Hope) is fun and justifiably well known. The canyon section is truly unique with good pavement, good scenery and plenty of curves. Unfortunately Yale to Hope isn't so special providing a dull ending to a great road. If you are experienced at night riding, consider taking the canyon at midnight. This is one of my favorite roads when ridden late on a hot summers evening.

You don't *have* to do the full loop. A turn-around at Lillooet is quite reasonable in my experience for about a 500-600km run. By Lytton or Cache Creek I usually prefer to continue on however.

The route feels quite different depending on the direction you choose so be sure to try it in both directions. Either way, it is one of BC's absolute best biking roads.

Agreed. I'd say it's an absolute must for the touring motorbiker - or, for that matter, the really, really well-conditioned touring bicyclist. Very little traffic once you get north of Whistler, too.

Anywhere in Vermont or New Hampshire is NUTS in the fall, because all the flatlanders from Mass come north to see the purdy colours and have to deal with life on a two-lane twisty road while looking at all the purdy colours.

The Ditch Grade just West of Redding Ca to 36 to 299 to the coast and back...or heck anywherre in Shasta, Trinity or Lassen area. All great twisty roads with little traffic and great sceanery. Just look out for cows and deer.

Given that we in MA "dog" about all the people who come from the south to see the colors - I doubt it's only MA people causing all the issues.

The Kancamagus Highway is great and all - but a few of the sideroads off of it were better, I found, for twistyness. The kanc is pretty, though.

To add a few Colorado rides to slimcolo's post:
125 North from Granby Colorado to Walden
Highway 14 from Walden to Fort Collins

And a really great loop is to drive over Trail ridge road (34) preferably in the morning when there is less traffic (just watch out for the wildlife) then depending on direction of travel, go North to Fort Collins and over Cameron Pass (highway 14) to 125 or the other direction. Highway 14 is probably my favorite drive in my car and I am planning on riding it on the bike for the first time next monday!
