Plugged hole in Carb 3

Ok, this hole is clogged in carb #3.

I don't have the special tool (brass bristle in a matchstick) to clear it out tonight. What are the possible problems caused by this? Could this be causing the bike to run rich? Could it prevent me from starting (actually working on carb #4, stopping for a break)?

I will come back and poke at it when I get a bristle and a matchstick, but for now I'm just gonna finish up the rest of the work.

You can generally blast blockages out here with pressure. If you're using an aerosol can cleaner take a small vaccum cap, poke a hole smaller than the cleaners straw through it, push the straw through and press the rubber cap against the hole. It allows the pressure to build up and should get rid of any blockage there. Won't help you with a really clogged jet but does the trick in most cases.

That won't make you run rich but it may prevent some flow and keep that cylinder from firing. I'm not familiar with your carbs and I don't know where that passage leads in the carb body

Make sure you get those jets and everything else clean. Be patient as a good job is better than having to pull the carbs again.

Guitar Strings can also be used like the "special tool" to clean passages. Just be carefull not to damage the holes or passages.
