Just bought a 2001 shadow spirit and I noticed a clunk noise when I shift into first. The remaining shifts dont sound noisey at all, is this normal?

Yes, especially when they're cold.

If you are going too fast when shifting down to first, the clunk will get louder. That tranny will feel most comfortable shifting down to first below around 10-15 mph.

I especially noticed that yesterday when I had to quickly gear down for a yellow light. The drop into 1st was very noticeable....click...click...CLANK!

Yeah, I get a cuh-clank kind of noise and feeling, sometimes my push to shift down isn't strong enough so it resists and I have to press harder. :P

Mine does the clunk as well unless I'm going very slow or stopped.
